Hassen Amri a donn\u00e9 le jeudi 31 mars 2022, une conf\u00e9rence intitul\u00e9e La r\u00e9trosynth\u00e8se organique pour la pr\u00e9paration de mol\u00e9cules bioactives et de m\u00e9dicaments dans le cadre de la chimie verte, la conf\u00e9rence a \u00e9t\u00e9 organis\u00e9e par le d\u00e9partement des sciences de l\u2019Acad\u00e9mie, et mod\u00e9r\u00e9e par la pr\u00e9sidente du d\u00e9partement, l\u2019acad\u00e9micienne Amel Benammar Elgaaied.<\/p>\n\n\n\n<p class=\"has-text-align-center has-vivid-red-color has-text-color\" style=\"font-size:25px\"><strong>R\u00e9sum\u00e9<\/strong><\/p>\n\n\n\n<p class=\"has-medium-font-size\">De nos jours, la chimie organique est tr\u00e8s sollicit\u00e9e parce qu\u2019elle est avant tout une science de cr\u00e9ation de mol\u00e9cules et de mat\u00e9riaux qui g\u00e9n\u00e8rent et fa\u00e7onnent beaucoup de supports vitaux (m\u00e9dicaments, cosm\u00e9tiques, polym\u00e8res, vernis, \u00e9lastom\u00e8res, fibres synth\u00e9tiques, r\u00e9sines, plastiques), pour ne citer que peu de beaucoup de d\u00e9riv\u00e9s de la chimie organique. Aussi parce qu\u2019elle est innovante, elle n\u00e9cessite du pratiquant une curiosit\u00e9, un esprit imaginatif, une rigueur et une pr\u00e9disposition pour appliquer les concepts de base n\u00e9cessaires \u00e0 la compr\u00e9hension et la proposition de nouvelles m\u00e9thodologies, pour d\u00e9velopper des synth\u00e8ses totales de produits organiques \u00e0 vertus diverses ou sans applications. La synth\u00e8se organique \u00e0 plusieurs \u00e9tapes demeure peu efficace en termes de faisabilit\u00e9, de rentabilit\u00e9, d\u2019optimisation mat\u00e9rielle et surtout peu garante du respect de l\u2019environnement. Elle a \u00e9t\u00e9 soutenue durant ces deux derni\u00e8res d\u00e9cennies par un nouveau concept, bas\u00e9 sur la technique de d\u00e9connexion des mol\u00e9cules finales ou cibles, en fragments fictifs dans l\u2019optique de leurs reconstructions en sens inverse ou vers l\u2019arri\u00e8re (1).<br>Cette pratique a \u00e9t\u00e9 initi\u00e9e par le chimiste Elias James Corey, p\u00e8re de la th\u00e9orie de la m\u00e9thodologie de synth\u00e8se organique et le concept de la r\u00e9trosynth\u00e8se,qui lui ont permis de produire le premier lot de prostaglandines en 1968et d\u2019\u00eatre r\u00e9compens\u00e9 par le prix Nobel de chimie en 1990 (2).<br>Compte tenu de la lenteur de la technique de la r\u00e9trosynth\u00e8se, chimistes et informaticiens ont con\u00e7u, gr\u00e2ce aux applications algorithmiques et l\u2019intelligence artificielle (IA), des logiciels performants dont le plus r\u00e9cent (5 fois plus rapide que l\u2019homme), est entr\u00e9 en activit\u00e9 en 2018 : Monte Carlo Tree Search (MCTS) (3).<br>Notre contribution dans le cadre de la synth\u00e8se de biomol\u00e9cules \u00e0 activit\u00e9s antibact\u00e9riennes ou anticanc\u00e9reuses par le biais dela r\u00e9trosynth\u00e8se organique, a \u00e9t\u00e9 bas\u00e9e sur des d\u00e9coupages demol\u00e9cules cibles en synthons th\u00e9oriques plurifonctionnels et la recherche de leurs \u00e9quivalents synth\u00e9tiques neutres, moyennant dessu bstrats organiques disponibles peu on\u00e9reux, des r\u00e9actifs basiques aqueux \u00e0 concentrations \u00e9lev\u00e9es tels que K2CO3 ou KHCO3, des amines tertiaires solubles dans l\u2019eau comme catalyseurs et le cas \u00e9ch\u00e9ant, peu de solvants organiques, en harmonie avec les principes de la chimie verte.<\/p>\n\n\n\n<p class=\"has-medium-font-size\">(1) S.
